With that slow twist is seams to duplicate the original Winchesters fairly close. The 50-96 was originally an express load loaded with a light bullet and a lot of black powder. The idea was to get more velocity over a longer range for point blank hunting. Mostly what I have read is that the 50-90 (96) uses bullets of around 300 gr. So you are going to need a light bullet to get accuracy. As for powder just fill the case with black powder leaving enough room to seat the bullet and compress the powder about 1/16 onch should get you started.

............I'd bet a large case like that (capacity) coupled with a short boolit is going to want some extra lube in the form of a lube cookie to help keep the fouling soft for follow-on accuracy. Like any other load work up, change only one thing at a time and keep good records of the pluses and negatives. Even the bad things are teaching you things.
