After a destroyed barrel in the late '80's, I quit fiddling with the Super concept, but I've delved fairly deep into what can be accomplished with a .45 pistol bullet. Even at hardball speed (830 fps) 230 grains can penetrate a lot - provided it doesn't expand:

That's a water-quenched wheelweight LBT 452-230-LFN recovered from it's ninth gallon water jug in the stack - about six feet of water and plastic. I'm not suggesting you should dial back from the +P+ / .45 Super velocity levels, but that you could stick with 230 grains in a flat point, have more room in the case for your propellant of choice, push it faster, have penetration in spades, and MAYBE get a little bit more hydrostatic damage (difficult to achieve in meat at ANY pistol velocity) than you would with a bigger 255 grain slug that's struggling with with the available case capacity and chamber space. My $0.02 anyway.
