Can someone tell me how a barrelmaker knows how deep the groove is that a single point cutter is making? I have have had excellent accuracy results with barrels made in this fashion and I am just curious as to how they measure the first and subsequent groove depth. I have a really poor shooting 280 Ruger 77 tang safety that I am pondering a re-barrel.

Never having done it, this is primarily guesswork:
1. Measure the height of the cutter to figure the depth of the groove.
2. Count the passes of the cutter through the barrel, while using the same number of shims or adjustments to the cutter.
3. If all else fails, slug the bore and compare depth of each groove.

I slug the barrel several times when rifling. If things are working right the cutter will only be taking out about .0002" per pass. A six groove barrel may take 120 passes.

No wonder they are more expensive. Do they need to be stress relieved after rifling is finished?

If the metal is stress relieved before machine work is done there should be no need to stress relieve again. Button rifling induces stress so the barrel needs to be stress relieved after.
