I picked up the old style lee hand primer with round trays and it is missing the cover for the large primer tray. I tried a cover off a press primer which fits, but I noticed this cover does not have a hole in it and the stock small primer cover does. So what is the hole in the cover for and do I need to drill one?

I've never used the hole on a hand primer.

I always used the small hole to add extra primers or to reinsert one I had to remove cause it was upside down. If your cover didn't have a hole, a drill would make it an easy fix.

YUP! That`s what the hole is for, although I rarely use it.Robert

I've missed the hole when I moved to priming tools without it, but not enough to do anything about it...yet

It is very handy for adding the odd primer that you had to remove back rather than putting it somewhere you might remember when done.
