Silly question but...
On the side of the lubrisizer where the actual die goes into, there is a hole. Lube comes out of this. I'm guessing this is to be plugged? Mine came with no plug so I was just making sure.

That's a bleed hole that needs to be plugged. Depending on the age of your machine, it can be plugged with either a screw plug or brass plug. I've got machines with both, but it does need to be plugged before pressure is applied to the lube.

In Andyc's picture, the smaller screw is a sizing die retainer, the bigger one drains the reservoir.
