I'm getting everything setup to start loading for my 1895 Cowboy. I've done a lot of reloading but most pistols so this is a little new. The load I'd like to start with is a 300gr RNFP over 45gr of 3031. I've done a lot of reading on here and I am not sure if I need a filler for this load, please help. Thanks

Sounds about right. You won't need any filler with that load. If you are using the Lyman reloading manuel or any of the others you should be fine. Elmer Keith used a 405 JSP over 50 grains of 3031 that was a real thumper on both ends. Stay with published data and you will fine.

No filler in a straight case.

I have been using 45gr Varget with Lee405HB with home make lube with good groups.
