Yes, if done correctly it would work no problem - in fact my experiments with developing loads for completely bio-degradable wadding (mostly Kraft brown paper based) have given me some indications that it is superior to plastic wadding components.
By "done correctly" I'm talking about using a nice round punch of the right diameter to punch out the over-powder/under-slug wads from heavy thick, non-hollow core, card-board which can be made from many layers of newspaper or brown paper with glue a flat surface and something big and flat to put on top of it while it dries. The paper tube to put around the slug should be rolled nice and tight around a rod or something similar from strips coated with glue and then slit like shot petals are slit.
In a TSHTF situation glue could be made from fine white flower and a little water.
Basically it would amount to making your own nitro cards punched from your own home made hard core thick card-board and rolling your own paper tubes. I already roll my own paper tubes since I have not been able to find a source of commercially available ones with the right inside and outside diameter with the one exception of some precision tubes from a “make your own fire-works” material wholesaler that sells some heavy well tubes that are the perfect size both internal and external diameter to make sabots for 20ga. rifled bore shotgun slugs that use 40-cal pistol bullets. I have to still cut them to length and slit them myself but it is less work then rolling my own tubes like every other combination requires. Obviously nitro-cards aren’t hard to get so I don’t make my own but I feel confident I could do so in a pinch if necessary.

Please can you detail what you do to convert 50BMG cases to shotshells......I thought 50BMG's were rimless while 12G's have a rim?.....

Wasp or hornets nest would be better for wadding. It does not smolder after firing and may prevent forest fires.
