Another vote for 8 to 8.5 grains of unique under either of those bullets. I shoot 9 grains under a 230 grain bullet for both rifle and pistol for hunting.

I can't add much - I have to echo what Hardcase416taylor said. That's the same load I finally settled on for my Uberti 45 Colt. I've used unique and GreenDot as well but still keep going back to the RD - the standard 255 gr. lead old time favorite flat nose. Can't speak to plated or copper clad but I would think that if you have Unique, it would certainly work with no problem.
I've never had an issue with the big volume 45 Colt casing and never felt the need for a card or filler of any type. With a standard large pistol primer, mine all go bang and shoot great.

I will add my vote for the 255gr/8.5 gr Unique load. It is my favorite load.
But a close second in my Vaquero, Blackhawk and Rossi 92 is the 255 gr bullet over 7.5 gr of Bullseye.
Unique a 250 to 255 grain bullet is probably been loaded in the 45 colt than any other combination and for a good reason they are accurate and fun. But if you have 4227 and no use for it and are worried about all the air space another fun accurate load is 20gr of 4227 and a 250 to 255gr cast bullet. If I recall correctly it is even safe to shoot in colts and the replicas. It is the preferred load of Hank Williams if that matters to you.
